Is there a reason(s) not to reuse drywall for a glass tile border?
Would the drywall need to be "primed"? with paint? something else? I removed a row of existing ceramic tile, along with the old mortar, from a counter-to-cabinet backsplash. Thx.

Depends on whether the tile will get wet.
If so, use either green board (available ar lumber yards), or better yet, use a concrete board - something like Hardy Plank.
If no water present, use the old sheet rock. No primer required.
