Older laptop with three USB ports managed to find and write software to a CH340 using Pyflasher. New laptop does the bing/bong as a CH340 is connected, but Pyflasher fails to show a com port, to enable the CH340 to be flashed.
I have tried known good USB leads for flashing, tried all three USB ports, tried turning off bluetooth - what else is worth trying?

I'd start by looking in device manager. The device should be there somewhere. If it shows up as "unknown device" then it doesn't have the drivers for it. If it shows up as a high-numbered COM port, the software might not like it - some software expects that COM ports only go up to COM4, while the actual limit I think is COM63. If that's the case, unplug the device, turn on "show hidden devices", and remove any COM ports. Then when you plug in again it should get COM3. Or of course if you have 60-odd COM ports already you'll need to delete them as you have no free numbers - Windows considers a device as "new" if either the serial number is different or (sometimes) if it's on a new USB port, so if you use a lot of devices they can accumulate, and it never forgets old ones.

I have managed to fix Windows Update by deleting the contents of \Windows\SoftwareDistribution as described in ttps://appuals.com/one-of-the-update-services-is-not-running-properly-in-windows-update/ near to the end of that page.
I am still working to resolve my original USB issue, the port is still flagged with a ! against it.
